The punchline is his choice of words and then he just decides to say fuck it and let the circle slide. I don't think there's any motif or deeper meaning beyond that. I found it funny, but I'm easily amused by stupid shit like this.

"Bi" means two. In the second frame she realizes that she's 29, which is odd and not even. The third frame is a knock down drag out fight over the definition of completeness in the real number set.

On a surface level reading, yes, that's the joke. But in a more subtle way (and this is what makes ST comics so clever) the second frame reveals to her (and by extension us as the audience) that she isn't just "bi"/"2", she is in fact 29 or "two nine" (which itself is a joke that is of course obvious enough to not need explaining) and that wrestling over the definitions of mathematical concepts is, in essence, to wrestle with oneself.
