Question. What tests are they using that can differentiate between the variants?

Mass-PCR using an Ion Torrent or similar machine. Basically using expensive equipment to run thousands of PCRs simultaneously.
RNA & DNA have their differences, but if you have equipment to read nucleotide base pairs (A, T, G/U, C) in DNA then you can read them in RNA. In such a case they would be looking for the genetic "signature" of the new variant.

Alternatively an Electron Micrograph, if they know the protein structure* of the new variant. This would be basically taking a nanoscopic 'photo' of sorts that can see the shape of the protein.

Proteins are made of 4 different levels:
1) Primary, this is literally just amino acid base pairs with maybe a few other molecules attached post-gensis.
2) Secondary, this is basically a ribbon made up of multiple strands of primary
3) Tertiary, which is multiple "ribbons" of secondary structure coiled, stacked, binded, etc. into different shapes. This is the building block structure.
4) Quaternary, This is the functional shape of the protein, anywhere from 1 - 30+ different tertiary blocks with the possibility of secondary ribbons/coils all built into something useful to whatever body is producing the protein.

When you hear people talk about the spike glycoprotein a.k.a "S-Protein" they are talking about the overall quaternary structure, which forms a specific suction cup of sorts optimized to bind onto specific receptors on the cell surface and allow entry of the viral capsid.
Now, because Nu Xi "Omicron" has significant changes to the S-protein, there are likely major tertiary building blocks in separate positions or outright using different tertiary blocks entirely.
If one knows which blocks are different/"out of place" (or rather "in place" given that the changes likely help the new variant), and you can successfully generate some kind of image or reading of the shape, you can isolate the structures and figure out if the protein fingerprint matches the new variant.

Oversimplified TL;DR: Proteins are like legos, if you can tell which pieces of the lego are built or placed different, than you can tell which lego models are which.

It makes sense that variants are becoming more out of control in areas with high vaccination rates. The virus would have to mutate more aggressively to stay alive and spread. There is a direct correlation, and vaccinations aren't going to stop it. If it can get a foothold anywhere, it's going to mutate to try to keep spreading.

I don't think vax companies editing their formula to compensate for Omicron is gonna do much. Holidays are coming and people are gonna be spreading it and all the vaccinated people should consider themselves just as at risk as they would be without a vaccine.
